This study compared two transglutaminase 2 (TG2) inhibitors, PX-12 and ERW1041, in celiac disease models. PX-12 was more effective at inhibiting purified and cellular TG2, but failed to inhibit extracellular TG2 on intact intestinal cells or reduce gliadin peptide transport, likely due to rapid degradation. ERW1041 consistently inhibited TG2 activity and significantly reduced transepithelial passage of the immunogenic gliadin peptide P56-88, supporting TG2 inhibition as a potential non-dietary therapy for celiac disease.
