This study demonstrates that vitamin D (VitD) levels and vitamin D receptor (VDR) expression correlate with histopathological differentiation in head and neck squamous cell carcinoma (HNC), with poorly differentiated tumors showing the lowest VitD levels and highest VDR expression. In vitro experiments show that VitD synergistically enhances cisplatin-mediated killing of HNC cells in both 2D and 3D tumor spheroid models, partly through downregulation of the PI3K/Akt/mTOR pathway. These findings suggest that VitD/VDR-targeted combination therapies may represent a promising, cost-effective strategy for improving treatment outcomes in HNC patients, particularly those with VitD deficiency.
