PROTECT-V is a platform trial testing pre-exposure prophylactic agents (intranasal niclosamide, inhaled/intranasal ciclesonide, and intravenous sotrovimab) against SARS-CoV-2 infection in immunocompromised and vulnerable patient populations. The trial uses a master protocol with shared placebo groups and adaptive features to evaluate multiple interventions efficiently across groups including transplant recipients, dialysis patients, and those with oncological, haematological, or autoimmune conditions. As of October 2022, 1611 patients had been randomised to the niclosamide arm and 68 to the sotrovimab arm, with the trial designed to address the ongoing unmet need for prophylaxis in patients with suboptimal vaccine responses.
