This study demonstrates that sialylation of conjunctival goblet cell mucins, primarily mediated by the sialyltransferase ST6GALNAC1, is essential for forming an impenetrable gel-like layer that encapsulates and removes allergens and particulate matter from the ocular surface. In a mouse model of allergic conjunctivitis, sialylated mucins significantly reduced clinical symptoms, eosinophil infiltration, and allergen uptake by dendritic cells. These findings reveal a critical protective mechanism of ocular surface mucins and suggest that enhancing mucin sialylation could be a therapeutic strategy for allergic eye diseases.
