This study demonstrates that viral transduction of a gain-of-function nicotinic acetylcholine receptor (α9L9′T) into cochlear hair cells of α9-knockout mice reduces temporary hearing loss after acoustic trauma. Treated mice showed smaller threshold shifts and faster recovery of auditory brain-stem response wave 1 amplitude compared to controls. These findings provide proof of principle for gene therapy to enhance efferent feedback as a complementary strategy for preventing noise-induced hearing loss.
