This study demonstrates that activation of D4 dopamine receptors inhibits Na+-K+-ATPase (NKA) activity in renal proximal tubule (RPT) cells from normotensive Wistar-Kyoto (WKY) rats via the NO/cGMP signaling pathway, but this effect is absent in spontaneously hypertensive rats (SHRs). The loss of D4 receptor function in SHRs is associated with reduced plasma membrane expression of the receptor despite increased total cellular expression. These findings suggest that impaired D4 receptor-mediated regulation of NKA activity in the kidney may contribute to the pathogenesis of hypertension.
