This study characterizes a CRISPR/Cas9-edited sorghum line (19Q4-13) with deletions in the alpha-kafirin gene family, resulting in a 43% increase in protein-bound lysine and a 161% increase in free lysine. RNA-seq and ATAC-seq identified 184 differentially expressed genes and 163 differentially accessible chromatin regions in developing kernels, with several changes conserved in maize prolamin mutants. The findings provide a resource for understanding proteome rebalancing and improving grain protein quality in sorghum.
