This study found that serum levels of pregnancy-specific glycoprotein 9 (PSG9) are significantly lower in women with preeclampsia compared to healthy pregnant women. In human umbilical vein endothelial cells, PSG9 at 0.1 μg/mL increased expression of Orai1 and Orai2 proteins, enhanced store-operated calcium entry, and promoted eNOS expression and nitric oxide production, effects that were blocked by the Orai inhibitor BTP2. These findings suggest PSG9 may regulate vascular endothelial function via a Ca²⁺/eNOS/NO signaling pathway and could have potential value in the prevention or treatment of preeclampsia.
