This study demonstrates that mitochondrial hydrogen peroxide (H₂O₂) produced early after partial hepatectomy (PHx) is essential for triggering liver regeneration in mice, rather than being merely a toxic byproduct. Scavenging mitochondrial H₂O₂ impaired regeneration, while NADPH oxidase inhibition had no effect. The regenerative signal is mediated through the Akt/Erk/FoxO3a/p27 signaling pathway, suggesting that improper antioxidant use could hinder liver recovery after surgery or injury.
