This retrospective study of 91 patients with advanced pancreatic ductal adenocarcinoma (PDAC) found that LV5FU2–carboplatin chemotherapy after failure of standard regimens yielded limited efficacy, with a median overall survival of 4.2 months and progression-free survival of 2.7 months. The clinical benefit rate was 25.2%, and the regimen showed acceptable tolerability with grade 3–4 neutropenia (24.7%) and thrombocytopenia (11.8%) as the most common severe adverse events. Clinically, this combination may be considered in selected patients without ascites or opioid-requiring pain, with longer disease evolution, and who are fit enough to receive full-dose carboplatin.
