Sun and colleagues demonstrate that the nuclear receptor LRH-1 directly induces asparagine synthetase (ASNS) and de novo asparagine synthesis in hepatocytes in response to acute liver injury, and this response limits hepatocellular damage. The hepatoprotective effect is mediated by asparagine itself, not by other amino acid metabolites, and does not affect hepatocyte proliferation. These findings suggest that asparagine supplementation could be a potential therapeutic strategy for acute hepatic injury.
