This study presents CRISPR-Clear, a method combining CRISPR-based knockout of the tyrosinase gene with PACT tissue clearing to enable deep optical imaging of melanin-rich B16 melanoma tumors. The approach eliminated melanin pigmentation without altering tumor cell morphology, gene expression, or in vivo growth characteristics, achieving a 10- to 30-fold increase in signal intensity at 300 µm imaging depth. This technique allows unprecedented optical access to intact melanoma tissue for antibody staining, nanoparticle imaging, and subcellular visualization, with potential applications in immuno-oncology and drug delivery research.
