This review examines how L-arginine and its methylated derivatives influence cancer cell proliferation and death. L-arginine deprivation selectively kills malignant cells while sparing normal cells, and D-arginine inhibits tumour growth, whereas L-arginine can stimulate tumour growth. The anti-proliferative and pro-apoptotic effects of methylated and hydroxymethylated arginine derivatives appear to be mediated through formaldehyde generation, suggesting potential applications in chemoprevention and cancer therapy.
