This study demonstrates that in pancreatic β cells under streptozotocin (STZ)-induced diabetic stress, Parkin-mediated mitophagy is activated via the Plk3-mtROS-PINK1-Parkin axis, leading to reduced cell growth and insulin secretion. FOXO3A acts as a negative regulator by inhibiting Plk3, thereby blocking mitophagy and protecting β-cell function. Antioxidants such as NAC and natural COA water also suppress this mitophagy pathway, suggesting potential therapeutic strategies for diabetes.
