This study investigates how the stimulatory G-protein alpha subunit (Gnas) in hypothalamic QPLOT neurons regulates daily metabolic rhythms in mice. Conditional deletion of Gnas in QPLOT neurons led to decreased nocturnal locomotion at 22°C and 28°C, and altered circadian patterns of energy expenditure, respiratory exchange, and food/water intake across different ambient temperatures. These findings advance understanding of Gαs-signaling in preoptic QPLOT neurons in regulating daily patterns of metabolism, with clinical significance for understanding central metabolic control and circadian rhythm disorders.
