This systematic review of 17 studies found that maternal folic acid supplementation during the preconception period and early pregnancy is associated with a protective effect against autism spectrum disorder (ASD) in offspring, with one study reporting a 40% reduction in risk. However, excessive folic acid intake, particularly from combined supplementation and food fortification, may be associated with increased ASD risk, suggesting a U-shaped relationship. The clinical significance is that while folic acid supplementation remains crucial for neural tube defect prevention, optimal dosing requires further investigation to balance benefits and potential risks for neurodevelopment.
