This study demonstrates that micromotion-induced fluid shear stress in the millipascal range promotes reactive gliosis around neural electrodes, mediated by mechanosensitive ion channels PIEZO1 and TRPA1. Using computational modeling, in vitro flow chamber experiments, and in vivo rat models, the authors show that shear stress upregulates gliosis markers and that chemical activation of PIEZO1 attenuates this response while inhibition exacerbates it. These findings identify mechanosensitive ion channels as potential therapeutic targets to improve long-term neuroelectrode performance.
