This study developed a novel gene therapy vector (scAAV2-Trx2-C3) that delivers a fusion protein of thioredoxin 2 and C3 transferase. In a dexamethasone-induced mouse model of glaucoma, a single intracameral injection lowered intraocular pressure for up to 4 weeks and protected retinal ganglion cells from apoptosis. These findings suggest a promising dual-action gene therapy approach for glaucoma that both reduces IOP and provides neuroprotection.
