This preclinical study combined behavioral data with untargeted metabolomics in rats to identify brain metabolites most relevant to acute MDMA-induced locomotor activity and those altered by repeated exposure. MDMA and its derivatives, serotonin, and dopamine/norepinephrine metabolites were the strongest predictors of acute behavior, while repeated exposure increased levels of specific MDMA metabolites.
