Twenty-eight pregnant F0 mice were randomized into four groups (n=7): control, ZnO NPs alone, ZnO NPs plus fresh orange juice (OJ), and OJ alone. Fifty percent of dams underwent cesarean section on gestational day 18 for pup evaluation; the remaining delivered naturally, and F1 offspring were examined at postnatal week 8. ZnO NP-exposed mothers and offspring showed significantly decreased body weight, increased liver weight, and elevated serum ALT, AST, and ALP levels compared to controls (p≤.05). Histopathological analysis revealed disrupted central veins, hepatocyte damage, pyknotic nuclei, and activated Kupffer cells in ZnO NP-exposed mothers and offspring. F1 pups exhibited morphological deviations including limb defects, open eyelids, and short tails. OJ co-administration ameliorated all measured toxicities. FRAP assays indicated reduced hepatic antioxidant capacity in NP-exposed groups.
